The perils of humanization – a mouse perspective

Summary: Let’s deep dive into the world of humanized mouse models! Over the last decade, the use of chimeric humanized mouse models engrafted with a variety of tissues and cells of human origin has become one of the experimental paradigms most frequently used in translational research. This has been possible thanks to the establishment of genetically engineered mice with severely impaired innate and adaptive immune systems, such as NSG or NOG mice, offering a highly permissive recipient environment for the functional integration of the xenotransplanted human components.
However, in the permissive and unregulated environment of the immunocompromised recipient mouse, the cellular interactions are sometimes unpredictable. In this context, a series of unintended post-transplant disorders have been characterized in recent years. These disorders are clinically relevant for mouse recipients, limiting their usage, especially for longer-term studies, and represent potential experimental confounders for interpreting preclinical data.
